چکیده

A bacterial isolate from soil samples contaminated with decaying lignocellulosic wastes and effluents of cotton industry was isolated for cellulase production and was identified as Bacillus subtilis subsp. inaquosorum after morphological, biochemical and phlogenetic analysis. Under optimized solid state fermentation conditions maximum cellulase production 71.15 IU/ml was obtained with wheat bran as carbon source in MA-1 mineral media and fructose and yeast extract as additives under 48 hr of production period at 37 ̊C, with 30% inoculum size. Moisture level of 1:2 (w/v) was found best for cellulase production. The crude cellulase so produced was further analysed for its potential applications in biostoning of denim. The purification of cellulase was carried out by ammonium sulfate precipitation followed by ion-exchange chromatography using DEAE-Sepharose and Sephadex G -150. The molecular weight of the purified carboxymethylcellulase (CMCase) was estimated to be about 32.5 kDa with the analysis of SDS-PAGE.

Cellulase Production Under Solid-State Fermentation by Ethanolic Zygomycetes Fungi: Application of Response Surface Methodology

Background and Objectives: Cellulase is an important enzyme with multiple applications in industries, including food, laundry, pharmaceutical, textile, pulp, paper and biofuel industries. Solid-state fermentation (SSF) is a method for cellulase production, which includes several advantages, compared to submerged fermentation. In this study, cellulase was produced by three filamentous fungi, i.e...

In the present study 10 bacterial isolates were isolated from the three different-different places soil sample by serial dilution and agar plate technique. These bacterial isolated colony were identified on the basis of colony morphology given by Bergey’smanual method and named as MJKS01 to MJKS10. Screening of the colony for the cellulolytic activity was done on the MAM (minimal agar medium) s...

Purification and biochemical properties of a thermostable, haloalkaline cellulase from Bacillus licheniformis AMF-07 and its application for hydrolysis of different cellulosic substrates to bioethanol production

A thermophilic strain AMF-07, hydrolyzing carboxymethylcellulose (CMC) was isolated from Kerman hot spring and was identified as Bacillus licheniformis based on 16S rRNA sequence homology. The carboxymethylcellulase (CMCase) enzyme produced by the B. licheniformis was purified by (NH4)2SO4 precipitation, ion exchange and gel filtration chromatography. The purified enzyme gave a single band on S...

Enhanced Production of Cellulase-free Xylanase by Alkalophilic Bacillus Subtilis Ash and Its Application in Biobleaching of Kraft Pulp

This paper reports high level production of a cellulase-free xylanase using wheat bran, a cost-effective substrate, under submerged fermentation by alkalophilic Bacillus subtilis ASH. Production of xylanase was observed even at alkaline pH up to 11.0 and temperature 60 °C, although the highest enzyme titer was recorded at neutral pH and 37 °C. The enzyme production under optimized fermentation ...
